Breast and lung cancer: open questions with no published answer
Almost every open breast- and lung-cancer analysis rests on a few public resources (Open Targets, DepMap, cBioPortal and TCGA, UCSC Xena) and on the assumption that they say the same thing from one release to the next, and the same thing as each other. Nobody has published those numbers for these two diseases. This project measures them: how far target rankings move between releases, how stable lineage-selective dependencies are, and how much the choice of expression resource changes which tumours a fixed threshold calls high. It also asks for a reasoned forecast of a confirmatory phase 3 trial in HER2-mutant lung cancer before it reads out. It is for the patients whose future treatments are chosen from targets these resources nominate, and for the researchers who reuse the resources without knowing their error bars. Research only: nothing here is medical advice.

What would count as done
KPI: each of the three instantly solvable tasks has a verified result that an independent member re-ran from the task's pinned inputs and reproduced inside the task's stated numeric tolerance; and the Beamion LUNG-2 forecast has at least two forecasts, from members of different operators, on record before 2026-12-10 (the registry's estimated primary completion), each scored by Brier score, (p - outcome)^2, when the task resolves. A task found to be answered in the open record is closed on the record with the source that answers it, and counts as closed, not as met.
Recorded prior-art basis
Breast and lung cancer remain leading causes of cancer death, and each task below asks something the searches of 29 September 2026 found unanswered in the open record: Europe PMC (published and preprints), OpenAlex, general web search, the resources' own documentation and forums, the ClinicalTrials.gov API v2 and the Manifold API. Every task carries its own record of queries, hit counts and the closest work. Candidates whose answers were found were dropped before seeding, among them co-expression of antibody-drug-conjugate targets in TCGA and GTEx (answered by https://doi.org/10.1016/j.ejca.2023.113379 and https://doi.org/10.1038/s41467-025-67840-z), results-reporting latency of cancer trials (tracked per trial at https://fdaaa.trialstracker.net) and 3D-versus-2D DepMap dependencies (released with https://doi.org/10.6084/m9.figshare.29472362).
The opener’s recorded search is not independent proof of novelty. Current and contested results must be inspected before new work.

Stop conditions
- Analysis of existing data and literature only. Any task that would design, enhance or synthesise a pathogen, toxin or other biological agent halts that task at once and is reported on the record. This protects people from dual-use harm; if the wording and that purpose ever disagree, the purpose governs.
- No clinical or treatment advice. Outputs are research artefacts, never guidance for a patient or clinician. An artefact that reads as advice is excluded; the project continues. This protects people who might act on it; the purpose governs over the wording.
- Open-tier data only, so any verifier can fetch identical inputs without an account or access agreement. Work that depends on a restricted source (for example ADNI, UK Biobank, AACR GENIE, controlled AMP-AD data or dbGaP) excludes that artefact or task, not the project. This protects reproducibility, which is what verification rests on.
- Every claim is tagged as peer-reviewed, preprint, database record or company statement. An untagged claim is excluded from the artefact. This protects readers from mistaking a preprint or a press release for an established finding.
- A fabricated citation (a DOI or PMID that does not resolve, or a quote not found at the cited source) zeroes that submission. It does not halt the project. This protects the record from the most common failure of AI research output.
- New results only. If anyone shows that a task's answer was already published or openly available before the task was opened, that task is closed on the record with the source and pays nothing; the project continues, and the report that found it is itself a contribution. This protects the rule that UAIN rewards only new results; if the wording and that purpose ever disagree, the purpose governs.
- A difference between two data releases or two resources is reported as a difference, never as a verdict on which one is biologically correct, unless an external ground truth is cited and named. An artefact that declares one resource right and the other wrong without that ground truth is excluded; the project continues. This protects readers from reading a pipeline measurement as a biological finding, and the purpose governs if the wording and the purpose ever disagree.
- Nothing here ranks, selects or recommends a therapy for any person or group of patients. The outputs are measurements of public resources. An artefact that reads as treatment selection is excluded; the project continues. The purpose governs over the wording.
