"claim_statement": "On primary evidence public by 2010-12-31, peginterferon plus ribavirin was the only approach with phase-3 randomized SVR24 evidence of durable cure: GT1 treatment-naive 42–52%, GT2/3 about 80%, prior nonresponders 7–18%, with SVR maintained in 99.1% over a mean 3.9 years. Adding telaprevir or boceprevir to it in genotype 1 was the best-supported near-term improvement, raising SVR24 in phase-2 RCTs: treatment-naive 54–75% in the arms significantly better than 38–46% controls; prior failures 51–53% vs 14%. No interferon-free, NS5A, cyclophilin, miR-122 or vaccine approach had shown durable cure; nitazoxanide had only small single-country GT4 SVR data.",

"note": "On 2026-09-25 I downloaded all 24 cited PubMed abstracts from NCBI efetch and checked each number in the claim against them. I checked the claim statement only. The detail field contains just a header, and the artifact T-28_synthesis.md is not retrievable through UAIN, so I have not audited the evidence matrix or the correction ledger.\n\nPegIFN/RBV:\n- Manns 2001 (PMID 11583749, Lancet): SVR24 genotype 1 (GT1) 42% in the higher-dose PegIFN arm; GT2/3 about 80% in every arm.\n- Fried 2002 (PMID 12324553): GT1 46%.\n- Hadziyannis 2004 (PMID 14996676): GT1 52% at 48 weeks with standard-dose ribavirin.\n- So \"GT1 naive 42–52%, GT2/3 about 80%\" holds.\n\nNonresponders:\n- Jensen REPEAT (PMID 19380853): arm SVRs of 16%, 7%, 14% and 9%.\n- Shiffman HALT-C lead-in (PMID 15057741): 18%.\n- Poynard EPIC3 (PMID 19208349): 14% in nonresponders.\n- So \"7–18%\" holds, taken as arm-level extremes.\n\nDurability: Swain 2010 (PMID 20637202, Gastroenterology, November 2010, before the cutoff) reports 99.1% of SVR patients with undetectable RNA over a mean 3.9 years. This comes from a follow-up cohort, not from the RCTs themselves, which the claim's wording allows.\n\nProtease inhibitors, treatment-naive:\n- PROVE1 (PMID 19403902): 61% (P=0.02) and 67% (P=0.002) vs 41%.\n- PROVE2 (PMID 19403903): T12PR24 69% (P=0.004) vs 46%. The other PROVE2 arms were not significant (T12PR12 60%, P=0.12), and the claim correctly leaves them out.\n- SPRINT-1 (PMID 20692693): 54%, 56%, 67% and 75%, all significant, vs 38%.\n- So \"54–75% in significant arms vs 38–46% controls\" is exact.\n\nProtease inhibitors, prior failures: PROVE3 (PMID 20375406) gives 51% and 53% vs 14%. Correct.\n\nNegative findings:\n- R1626 (PMID 18570306, plus the week-72 letter PMID 18821613): only week-4 RNA and a high relapse rate.\n- INFORM-1 (PMID 20951424): all-oral, 14-day RNA change only.\n- BMS-790052 NS5A (PMID 20410884): single-dose RNA decline.\n- Debio-025 (PMIDs 18302285, 19353740): 4-week RNA only.\n- miR-122 (PMID 19965718): chimpanzees.\n- IC41 vaccine (PMIDs 18471515, 19559112): T-cell and RNA responses only.\n- Nitazoxanide: Rossignol, Egypt, GT4 (PMIDs 19135998, 18616643): 23 to 28 per arm, SVR 17–79%.\n- So \"no durable cure; nitazoxanide only small single-country GT4 SVR data\" holds.\n\nMinor caveat, not a reason to dispute: standard IFN alfa + RBV also had phase-3 SVR evidence before PegIFN. \"Only approach\" is exact if IFN-based therapy is read as one category, which is how the task frames it.",

Its detail contains only an introductory paragraph: the advertised matrix and reasoning are absent. RC-4543 explicitly supplies a replacement, but I do not certify that separate submission here. I independently retrieved NCBI efetch XML abstracts and publication dates for PMIDs 11583749,12324553,14996676,19625712,20637202,19380853,15057741,19208349,19403902,19403903,20375406,20692693,18616643,19135998. All are pre-2011. This is a partial reproduction of the claim, not a complete audit of all listed sources or the unavailable artifact. Reproduced: Manns GT1 42%, GT2/3 about 80%; Fried GT1 46%; Hadziyannis GT1 52%; Swain 99.1% over mean 3.9 years (cohort includes peginterferon monotherapy as well as combination); REPEAT 7/9/14/16%; HALT-C 18%; EPIC3 nonresponders 14%. PROVE1 61/67 vs41, PROVE2 significant triple arm69 vs46, SPRINT1 54/56/67/75 vs38, PROVE3 triple51/53 vs14 reproduce the PI ranges. Caveats prevent confirming the exact claim: (1) GT1 42–52% omits IDEAL (PMID19625712, Aug2009; n3070) with 39.8%,38.0%,40.9%; even standard doses give values below42. (2) HALT-C's18% is from a common-treatment retreatment lead-in, not a randomized comparison of retreatment regimens; separate it from randomized REPEAT7–16%. (3) Nitazoxanide monotherapy (PMID18616643, Sep2008) measured SVR24 in4/23, so the unqualified interferon-free negative requires qualification. This small study does not establish multi-year durability, but neither may its measured SVR24 be erased. Restrict the negative to interferon-free DAA combinations or explicitly distinguish SVR24 from long-term follow-up. (4) Literally 'only approach' is too strong: Fried's randomized conventional IFN/RBV and peginterferon-alone arms also measured SVR24; Swain includes166 peginterferon-alone recipients. Say best-established combination rather than only approach. These are claim/method limitations, not project-scope exclusions. No artifact is excluded.",
