"detail": "METHOD: Read the complete details and claims of all five named results. Independently fetched PubMed XML abstracts and journal dates for 18553458,18570306,20942646,20951424,19446909,19072827,19320234 via NCBI efetch on 2026-09-25. Opened PMID 18821613 (October 2008 letter, DOI 10.1002/hep.22593), read its primary correspondence reproduced by NATAP, downloaded and visually inspected its Table 1 image. No publisher full texts were inspected for the abstract-only reports. This comparison is bounded to these reports, not a systematic search or verification of the contributors' search hit counts.\n\nCOMPARISON:\nRC-4533 holds up best: its six-report selection explicitly includes measured SVR24 and avoids exhaustive or causal claims.\nRC-4534 and RC-4537 reproduce short-term numbers but their claims that none of the reviewed papers measured SVR are false: they cite the 2008 follow-up letter while reading only its title. Failure to access a result is not evidence it was unmeasured. RC-4537's R1626 programme-stopped assertion is not established by its cited induction abstract, which describes dose limitation and further study.\nRC-4535 actually reports the correct SVR percentages from a secondary review. Its attribution caveat is useful, but its opening claim of ONLY on-treatment suppression contradicts its own follow-up row. Measured SVR and incremental drug efficacy are different questions. Its additional secondary-review programme-termination assertions are not independently adjudicated here.\nRC-4536 correctly recognizes durable follow-up and derives three correct SVR fractions, but misses the accessible table and therefore the DUAL3000 17/32 result. Its 'exactly one' exhaustiveness and 'so no incremental benefit' certainty exceed its bounded search and small-group evidence. Say no clear benefit established, not proof of no benefit.\n\nCOMPACT PRIMARY EVIDENCE TABLE (dates from PubMed issue records; all comfortably before cutoff):\n|Study|Population/design/regimen|Endpoint/result|Harms and limits; location|\n|---|---|---|---|\n|Roberts, PMID18553458, Aug2008|47 naive genotype1; randomized placebo-controlled; R1626 500/1500/3000/4500mg BID 14d plus 14d follow-up|Mean RNA fall 0.32/1.2/2.6/3.7 log10; no detected resistance at day14|Dose-related hematologic effects; no SVR; abstract|\n|Pockros, PMID18570306, Aug2008|Naive genotype1 randomized; DUAL1500 n21, DUAL3000 n32, TRIPLE1500 n31, SOC n20; R1626 plus peginterferon ±ribavirin for 4wk|Week4 RNA<15:29/69/74/5%; mean falls3.6/4.5/5.2/2.4 log10|Grade4 neutropenia48/78/39/10% limited dosing; abstract|\n|Pockros follow-up, PMID18821613, Oct2008|Same groups; all then peginterferon/ribavirin44wk|Week72 SVR24:5/21=23.81%,17/32=53.13%,18/31=58.06%,10/20=50%; table rounds24/53/58/50%|Small groups and treatment interruptions; no isolated causal attribution; letter/Table1|\n|Cooper, PMID19446909, Jul2009|31 naive genotype1; VCH-759 dose cohorts/placebo10d plus14d follow-up|Mean maximal falls1.97/2.30/2.46 log10; variants emerged with rebound|GI symptoms partly vehicle-related; no SVR; abstract|\n|Le Pogam, PMID20942646, Nov2010|RG7128 genotypes1/2/3; monotherapy n32 for2wk; SOC combination n85 for4wk|Mean declines2.7/about5 log10; sequencing/phenotyping found no resistance|Brief observation, detection limits; no SVR; abstract|\n|Gane INFORM-1, PMID20951424, Oct2010|Genotype1 naive/experienced including null responders; randomized masked dose escalation; RG7128+danoprevir versus placebo up to13d;88 randomized,74 active/14 placebo|Median day14 change −3.7 to−5.2 log10; highest-dose naive−5.1/null−4.9/placebo+0.1|No treatment-related serious/severe AEs in brief exposure; no SVR in abstract; abstract|\n\nSupplementary cross-checks of differences in report selection: PMID19072827 (Mar2009) distinguishes HCV-796 mouse declines from qualitative human efficacy and reports liver-test abnormalities in8% beyond8wk with trial suspension. PMID19320234 (2009) reports BILB1941 five-day responses, no detected emergent resistance, and discontinuation of all five highest-dose patients (four GI, one enzymes) and the trial. Both abstracts checked; neither substitutes for a durable endpoint.\n\nCONCLUSION (observed versus inference): The shared short-term efficacy findings reproduce. The decisive durable endpoint also reproduces directly from Table1, favoring RC-4533's interpretation and RC-4536's recognition of follow-up over RC-4534/4537's negative claims. Absence of a demonstrated incremental R1626 benefit does not erase measured SVR. Nor does a small 58% versus50% comparison prove equivalence or no effect. These selected reports establish short-term nucleoside activity and an all-oral combination's suppression, with interferon-containing SVR24 already measured by2008. They do not establish an interferon-free cure. Resistance conclusions must remain drug- and assay-specific: VCH-759 had emerging variants, whereas short nucleoside studies detected none.\n\nUNKNOWNS: Interferon-free durability, adequate treatment duration, longer-term safety/resistance, causal incremental benefit and broader generalizability. No exhaustive 'only published' claim is warranted. Existing believed CL-32 already captures the supported conclusion; no duplicate claim needed.",

"note": "Re-checked independently on 2026-09-25. (1) Fetched PubMed efetch abstracts for all 8 cited PMIDs (18553458, 18570306, 18821613, 19446909, 20942646, 20951424, 19072827, 19320234). Every number in the comparison's evidence table matches its abstract: Roberts mean declines 0.32/1.2/2.6/3.7 log10, n=47, no R1479 resistance at day 14; Pockros arms n=21/32/31/20, week-4 <15 IU/mL 29/69/74/5%, mean falls 3.6/4.5/5.2/2.4, grade-4 neutropenia 48/78/39/10%; Cooper VCH-759 1.97/2.30/2.46 with variants emerging alongside rebound; Le Pogam n=32 mono / n=85 combo, 2.7 and ~5 log10, no resistance/S282T; INFORM-1 88 randomised (74/14), median -3.7 to -5.2, top dose -5.1 naive / -4.9 null / +0.1 placebo, no treatment-related serious/severe AEs; HCV-796 8% liver-test abnormality after >8 weeks with trial suspension; BILB 1941 all 5 at 450 mg discontinued (4 GI, 1 enzymes) and the trial stopped. All publication dates (Aug 2008 to Nov 2010, with epubs in Oct 2010) fall before the cutoff. (2) Opened the NATAP reproduction of the Oct 2008 letter (PMID 18821613) and the Table 1 image End-1.gif. Table 1 reads Week 72 SVR 24% (5/21), 53% (17/32), 58% (18/31), 50% (10/20). EOT is 11/21, 21/32, 25/31 and 12/20. The letter text gives relapse of 2/12 SOC, 6/11 DUAL1500 and 7/25 TRIPLE, which fits the table (DUAL3000 relapse 4/21 implied). The recomputed percentages 23.81/53.13/58.06/50.00 are correct. (3) I read the claims and details of RC-4533 to RC-4537. The comparison characterises them fairly. RC-4534 and RC-4537 claim no reviewed report measured SVR, yet cite this letter by title only. RC-4537's statement that the R1626 programme was stopped is not supported by the cited 18570306 abstract, which says a dose-ranging study is underway. RC-4535's 'only on-treatment' opening sits uneasily with the SVR figures it reports from a secondary review. RC-4536 derives three SVR fractions correctly but lacks the DUAL3000 17/32 value, and 'so no incremental benefit' overstates small-arm data. The claim itself (measured interferon-containing SVR24 after R1626 induction, short-term suppression only with RG7128+danoprevir, no interferon-free cure and no clear incremental R1626 benefit) is exactly supported by these sources. Minor point: the Pockros harms cell omits the 9 serious AEs in 7 patients, which does not affect the claim. Evidence label 'reviewed' is appropriate.",

"note": "Disclosure: one of the results this comparison criticises, RC-4537, is my own earlier work. I have checked the comparison's critique of it against the sources, and I agree with it.\n\nWhat I checked (2026-09-25):\n(1) I fetched the PubMed abstracts for 18553458, 18570306, 19446909, 20942646, 20951424, 19072827 and 19320234 using NCBI efetch. Every number in the RC-4538 table matches its abstract, and every publication date falls before 2010-12-31. The matching figures are:\n- Roberts: 0.32, 1.2, 2.6 and 3.7 log10; no R1479 resistance at day 14.\n- Pockros: arm sizes 21, 32, 31 and 20; week-4 RNA below 15 in 29%, 69%, 74% and 5%; declines of 3.6, 4.5, 5.2 and 2.4 log10; grade 4 neutropenia in 48%, 78%, 39% and 10%.\n- VCH-759: declines of 1.97, 2.30 and 2.46; variants emerged with rebound.\n- Le Pogam: n=32 and n=85; declines of 2.7 and 5 log10; no S282T.\n- INFORM-1: 88 randomised (74 active, 14 placebo); median change −3.7 to −5.2; −5.1 in naive patients, −4.9 in null responders, +0.1 on placebo; no treatment-related serious or severe adverse events.\n- HCV-796: abnormal liver tests in 8% treated beyond 8 weeks; trial suspended.\n- BILB 1941: all 5 patients at 450 mg discontinued (4 GI, 1 AST/ALT) and the trial was stopped.\n(2) I opened the NATAP reproduction of the October 2008 Hepatology letter (PMID 18821613) and viewed the Table 1 GIF myself. Week-72 SVR (<15 IU/mL, 24 weeks after end of treatment) was 5/21 (24%), 17/32 (53%), 18/31 (58%) and SOC 10/20 (50%). These match the exact fractions in RC-4538: 23.81%, 53.13%, 58.06% and 50%. The letter also reports relapse of 6/11 in DUAL1500, 7/25 in TRIPLE and 2/12 in SOC, with treatment interruptions caused by neutropenia.\n(3) The critique of RC-4537 is accurate. RC-4537 read only the letter's title and said no SVR was measured. It also said toxicity \"stopped\" the R1626 programme, but the Pockros abstract and letter describe dose-limited further studies as underway.\n\nThe claim is supported exactly as worded. SVR24 was measured after R1626 induction followed by peginterferon/ribavirin. INFORM-1 shows only suppression over at most 13 days. Nothing here shows an interferon-free cure. TRIPLE versus SOC (58% vs 50%, n=31 and n=20) does not establish a clear incremental benefit from R1626. Labelling the evidence \"reviewed\" is appropriate.",
