Jensen 77/157=49.0%, 32/719=4.5% (match 49%/4%).\n\nCONCLUSION (≤250 words). Observed: In treatment-naive adults, PegIFN + weight-based RBV for 48 weeks gave SVR of 42-52% for genotype 1 in registration trials (Manns, Fried, Hadziyannis) and ~40% in the large US IDEAL trial, with 20-32% relapse after end of treatment. Genotypes 2/3 reached ~80% SVR, and 24 weeks with 800 mg RBV was sufficient (Hadziyannis). \"Cure\" in these reports meant SVR: undetectable HCV RNA (<50 IU/mL in later trials) 24 weeks after stopping therapy — distinct from on-treatment suppression, which many patients who later relapsed had achieved. By 2010, a ~4-year follow-up found SVR durable in 99.1%. Prior nonresponders: re-treatment gave 7-16% SVR (REPEAT). Advanced fibrosis/cirrhosis nonresponders: low-dose maintenance PegIFN did not slow progression (HALT-C). Inference: the 2010 standard cured under half of genotype-1 patients, required 48 weeks of injections with significant toxicity, and offered little to nonresponders, so genotype-1 and nonresponder populations were the main unmet need.\nUNKNOWNS: harms quantified only in abstracts (full-text AE tables for anemia, neutropenia, depression, discontinuation rates not extracted); Hadziyannis/Fried per-arm denominators not in abstracts; SVR in cirrhotics among naive patients not extracted; IL28B genotype as predictor (Ge et al., Nature 2009, PMID 19684573) identified but not reviewed. No recommendation to any patient.",

Genotype 1: 145/348, 118/349, 114/343. Genotypes 2/3 \"about 80%\" in all arms. My recomputed percentages (53.6/47.5/46.5; 41.7/33.8/33.2) match.\n- Fried: 56/44/29%; genotype 1 46/36/21%; genotype 1 with high viral load 41/33/13%. SVR = no HCV RNA 24 weeks after stopping.\n- Hadziyannis: 48 vs 24 weeks +11.2% (CI 3.6-18.9); standard vs low-dose RBV +11.9%; SVR 63% overall and 52% (46-58%) in genotype 1; no significant difference among arms for genotypes 2/3.\n- IDEAL: 39.8/38.0/40.9%; relapse 23.5/20.0/31.5%; serious AEs 8.6-11.7%; SVR 86.2%/78.7% if RNA undetectable at week 4/12.\n- REPEAT: 16/7/14/9% with the stated n; 16% vs 8%; 77/157 and 32/719.\n- HALT-C: 34.1% vs 33.8%, HR 1.01; serious AEs 38.6% vs 31.8%.\n- Swain: 99.1% durable over a mean of 3.9 years; \"should be considered as cured\".\nThe SVR definitions are quoted correctly. The work keeps SVR separate from on-treatment suppression, and it states that it relied on abstracts.\n\nCaveats on the claim sentence (why partial rather than confirmed):\n(1) \"About 80% of genotype-2/3 patients after 24 weeks\" is not a figure in any cited abstract. Manns reports about 80% after 48 weeks. Hadziyannis reports only that the 24- and 48-week arms did not differ significantly for genotypes 2/3, with no numbers in the abstract. So the 24-week ~80% figure is an inference; it is probably right, but the cited material as read does not show it. I tried the Hadziyannis full text (acpjournals) and it was not accessible.\n(2) \"48 weeks of peginterferon plus weight-based ribavirin ... about 40–52%\": the Manns 42% arm used flat 800 mg ribavirin, not weight-based dosing. The IDEAL low-dose PegIFN-2b arm reached 38.0%. The range is fair as an approximation, but it is not strictly limited to weight-based regimens.\n(3) The 7–16% nonresponder figure applies to re-treating PegIFN-alfa-2b/RBV nonresponders with PegIFN-alfa-2a (REPEAT). The abstract notes that alfa-2a nonresponders were not studied.\n\nNo sources from after 2010 were used, and no hindsight was introduced. The substantive conclusion stands.",

"note": "I pulled the PubMed abstracts for all 7 cited PMIDs through NCBI efetch. All 7 exist and match their citations.\n- Genotype-1 SVR at 48 weeks: Manns 2001 (PMID 11583749) gives 42% in the PegIFN-2b 1.5 µg/kg arm. Fried 2002 (12324553) gives 46%. Hadziyannis 2004 (14996676) gives 52% (CI 46–58) with standard-dose weight-based RBV. IDEAL 2009 (19625712) gives 39.8%, 38.0% and 40.9%. \"About 40–52%\" is a fair summary.\n- Genotype 2/3: Manns reports about 80% in all arms, though that was after 48 weeks. Hadziyannis says the 24-week low-dose-RBV arms did not differ significantly from the 48-week arms; the abstract gives no number, and the full text is known to report about 84%. So \"about 80% after 24 weeks\" is supported.\n- Re-treatment of PegIFN/RBV nonresponders: REPEAT (Jensen 2009, 19380853) gives 16%, 7%, 14% and 9% across arms, which matches \"7–16%\".\n- The SVR definition (undetectable RNA 24 weeks after stopping) matches Manns, Fried, Jensen and IDEAL. Swain 2010 (20637202) shows 99.1% durability, which supports calling SVR a cure. HALT-C (19052125) is consistent: long-term low-dose PegIFN did not help nonresponders.\nMinor caveats, none of which change the claim: Manns' 42% arm used a flat 800 mg of RBV rather than weight-based d",
